member of the Bcl-2 family of proteins. During
mitochondria-regulated programmed cell death, Bak
promotes apoptosis by binding Bcl-2 a n d inhibiting
its anti-apoptotic function (1). Upon apoptotic
stimulation, Bak translocates from the cytosol to
the mitochondrial membrane (2). Pro-apoptotic
cascade activates BID, which oligomerizes Bak
into p o r es that result in the release of
cytochrome c (2, 3). In healthy cells, Bak associate
with Bcl-xL a n d Mcl-1 but not with Bcl-2,
Bcl-w o r A1 a n d only when freed of both Bak will
induce apoptosis.
