as SLP65, BASH, a n d BCA) is a B cell adapt o r that
plays an imp o r tant role in B cell development a n d
in BCR-signal transduction. Since BLNK does not encode
any intrinsic enzymatic activity, its function is to
serve as a scaffold f o r assembling
macromolecular complexes which includes enzymes
(PLC gamma, Vav a n d Btk) a n d additional linker
proteins (Grb2 a n d Nck) (1-3) . In addition, BLNK
binds Btk a n d is required f o r activation of
the transcription fact o r NF-κB. Tyrosine phosph
o r ylation of BLNK by Syk provides specific docking sites
f o r these SH2-containing effect o r proteins which in
turn permits phosph o r ylation a n d / o r activation
of their respective signaling pathways.
