expressed protein found in numerous actin structures.
It consists of an N-terminal actin binding domain, a
central rod domain, a n d a C-terminal domain
a n d functions as a homodimer to cross-link
actin filaments. The rod domain determines the
distance between cross-linked actin filaments a n d
also serves as an interaction site f o r
several cytoskeletal a n d signaling proteins (1).
Alpha-actinins are a multigene family of four
actin-binding proteins related to dystrophin. The
two skeletal muscle isof o r ms of alpha-actinin
(ACTN2 a n d ACTN3) are maj o r structural components
of the Z-line involved in anch o r ing the
actin-containing thin filaments. In humans, ACTN2
is expressed in all muscle fibers, while ACTN3 expression
is restricted to a subset of type 2 fibers (2).
