(JAK) tyrosine kinases involved in cytokine
receptor-mediated intracellular signal transduction (1).
The JAK family is constitutively associated with
cytokine receptors; upon ligand binding, JAK
phosphorylates the receptor. JAK also phosphorylates
the cytokine receptor on tyrosine (Tyr) residues that
serve as docking sites for cytoplasmic signal
transducers and activators of transcription (STAT)
proteins (2). Activated STATs translocate to the nucleus
to mediate gene transcription (1, 3). JAK2 is
activated after binding to the following
Interleukin receptors: specifically IL-3 and IL-23 (4).
It also plays a role in leptin signaling and control
of body weight (4). A chromosomal aberration involving
JAK2 is found in a form of pre-B acute myeloid leukemia (5).
