tyrosine kinases (Jaks), whose function is to
transduce extracellular signals from cytokines
a n d interferons. Jaks associate with cytokine recept
o r s; upon lig a n d binding, Jaks phosph o r ylate
the recept o r . The phosph o r ylated recept o r s
are bound by SH2-containing proteins, one class of which
is the signal transducers a n d activat o r s
of transcription (STATs). Activated STATs translocate to
the nucleus to mediate gene transcription (1, 2). Jak3
is activated after binding to the gamma chain of
Interleukin recept o r s, specifically IL-2, IL-4,
IL-7, IL-9, a n d IL-15 recept o r s. Jak3 has
been implicated in mast cell-mediated
immediate hypersensitivity reactions, autoimmune dis
o r ders, a n d acute lymphoblastic leukemia. Jak3 inhibit
o r has shown clinical potential as a new class
of immunomodulat o r y agents with anti-leukemic
a n d immunosuppressive properties (3).
