a 170 kDa transmembrane glycoprotein recept o r
tyrosine kinase that, activated by epidermal growth fact
o r (EGF), affects cell growth a n d differentiation
(1). Binding of EGF o r TGF alpha to EGFR activates
tyrosine kinase activity of the recept o r (2). The
carboxy terminal tyrosine residues on EGFR, Tyr 1068,
Tyr 1148, a n d Tyr 1173, are the maj o r sites
of autophosph o r ylation, which occurs as a result of
EGF binding (3). Once activated, EGFR
phosphotyrosines Y-1068 a n d Y-1173 mediate the binding
of Grb2 to the EGFR. Another autophosph o r ylation
site, Tyr 1086, is situated within the cytoplasmic domain,
a n d is phosph o r ylated to a higher extent by c-Src
than by EGFR (4).
